【Abstract】 AIM: To evaluate the inhibitive effects of Astragalus and Thoroughfare decoction against hepatitis B virus(HBV) in vitro. METHODS: HepG2.2.15 cells, as target cells, were exposed to different concentrations of Interferon α (IFNα) or Astragalus and Thoroughfare decoction respectively. Realtime PCR was applied to quantify extracellular HBV DNA and enzymelinked immunosorbent assay (ELISA) was used to assess HBsAg and HBeAg. RESULTS: The 50% inhibitory concentration (IC50) of Astragalus and Thoroughfare decoction against HBsAg, HBeAg, and extracellular HBV DNA was 1.35, 1.92 and 2.19 g/L respectively, and treatment index (TI)>3.56-5.79. αIFN had weak inhibitory effects against HBV. CONCLUSION: Astragalus and Thoroughfare decoction can inhibit HBV replication in vitro to some extent.

【摘要】 目的: 观察黄芪蛰虫合剂体外抗乙型肝炎病毒 (HBV) 的作用. 方法 : 以HepG2.2.15细胞为靶细胞,分别加入不同浓度的黄芪蛰虫合剂和α干扰素. 采用ELISA法及荧光定量PCR法,测定药物对细胞培养上清HBsAg, HBeAg及细胞外HBV DNA的抑制作用. 结果: 黄芪蛰虫合剂具有一定程度的抗HBV的作用,其抑制HBsAg, HBeAg及细胞外HBV DNA的50 %抑制浓度(IC50)分别为1.35, 1.92, 2.19 g/L. 治疗 指数(TI)>3.56~5.79. αIFN也有抑制HBV的作用……
