人骨髓间充质干细胞对K562细胞增殖和化疗敏感性的影响药学 |

【摘要】 本 研究 比较K562细胞与正常人骨髓间充质干细胞(MSC)黏附培养前后细胞增殖、化疗敏感性及MDR1变化,以寻找白血病耐药与造血微环境的关系。对悬浮培养和与MSC黏附培养的K562细胞绘制细胞生长曲线;用流式细胞术测定细胞周期并观察化学药物对细胞存活率和凋亡的 影响 ;用RT-PCR技术检测MDR1基因表达。结果表明:与悬浮培养比较,黏附培养K562细胞增殖受抑,G0/G1期细胞比例增加 (P<0.05),S期细胞比例下降 (P<0.05),G2/M期细胞比例增加(P<0.01)。在柔红霉素(DNR)诱导的凋亡中,黏附培养组K562细胞凋亡受阻 (P<0.05)。黏附培养未诱导K562细胞MDR1基因表达,也未使K562/ADM细胞的MDR1基因表达发生改变。结论: K562细胞与MSC黏附接触共培养,可导致K562细胞生长抑制,并产生化疗耐药,其机制可能与MDR1无关。

Influence of Human Mesenchymal Stem Cells on Cell Proliferation and Chemo-sensitivity of K562 Cells
Abstract This study was aimed to compare K562 cell proliferation,chemo-sensitivity and alteration of MDR1 before and after adhesive culture with MSC,so as to evaluate the relationship between chemodrug-resistance of leukemia cells and hemopoietic microenvironment. K562 cell cultivated in suspension and adhesively cultivated with MSC were collected respectively and cell proliferation curves were drawn; the cell cycle was determined by flow cytometry; the effect of chemotherapy on cellular viability and apoptosis of K562 cell was investigated,the MDR1 gene expression……
