Ageing lens and development of cataract
【Abstract】 The structural changes of the ageing lens include the increase of lens mass and lens density, the thickening of the anterior lens capsule and the decrease in density of epithelium. The accumulation of fluorogen may play a major role in the yellowing and increasing fluorescence of the lens with age. Increased posttranslational modification of proteins and unfolding of proteins eventually result in the aggregation of proteins and the inactivation of enzymes. The powerful antioxidant scavengers and chaperone molecules of lens are minimized during ageing, thus aggravating the damage induced by posttranslational modification. Cataract, the final stage of the ageing lens, may be regarded as a signal of ageing in other organs. Molecular chaperone may rescue the conformational diseases and revive the inactive enzymes in lens.

【摘要】 晶状体老化的结构性改变包括晶状体质量和密度的增加、晶状体前囊膜的增厚和晶状体上皮细胞密度的降低等.荧光发生素在晶状体中积累,可能主要参与老化过程中晶状体变黄和荧光色素增加.老化晶状体中蛋白质翻译后修饰和蛋白质去折叠增加,最终导致蛋白质凝聚和酶失活.晶状体特有的氧化清除剂和分子伴侣作为强大的防御系统,在老化过程中逐渐衰退,加剧了翻译后修饰造成的损伤.白内障是晶状体老化的最终结果,可作为其他组织器官衰变的信号.分子伴侣能解救构象性疾病,使晶状体重要的代谢酶‘起死回生’.
