【摘要】 目的 探讨褪黑素(melatonin,Mel)对大鼠缺血再灌注(IR)损伤的保护作用及机制。方法 通过手术建立大鼠肾缺血再灌注损伤模型,将实验动物随机分为Sham组、IR组和Mel+IR组,测IR后24 h各组大鼠血清中尿素氮(BUN)和肌酐(Cr)、肾组织匀浆中超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量变化,并观察肾脏组织结构的病理变化。结果 IR后24 h大鼠血清BUN、Cr水平明显升高,组织匀浆SOD活力明显降低,MDA水平明显升高。Mel+IR组较IR组肾脏SOD活性明显升高(P<0.01),MDA水平明显降低(P<0.01);肾功能、组织变化较IR组得到了明显改善。结论 褪黑素可通过提高组织抗氧化酶活性、降低组织脂质过氧化反应发挥对肾功能及肾脏结构的保护作用。

Abstract Objective To investigate the protective effect and mechanism of melatonin on renal ischemia/reperfusion(IR) injury in rat . Methods Using an in vivo rat model of IR, we observed the change of BUN and Cr in blood serum, and changes of the activities of superoxide dismutase (SOD), and malondiadehyde (MDA) in homogenized kidney tissue, and tissue structural pathological change during 24h after reperfusion.
